The Effects of Soamgudamikgitang on the Immuno-modulation and Gap Junctional Intercellular Communication |

ABSTRACT |
Objective : In order to observe the effects of SAKT on anticancer properties and GJIC, we have measured cytotoxicity on WB-F344, restoring effects of inhibited GJIC and Immuno-modulation effect of SAKT.
Method: 1. Cytotoxicity of SAKT on WB-F344 was measured by MTT assay. 2. WB-F344 was processed with H2O2 and SAKT 200㎍/㎖, 400㎍/㎖ , and 800㎍/㎖ respectively. Transfering pattern of lucifer yellow was observed by Scrape loading dye transfer (SL/DT) assay. 3. Experimental mice were divided to sarcoma-180 injection group (control), sarcoma-180 with SAKT 10mg injection group(SAKT 10), and sarcoma-180 with SAKT 50mg injection group(SAKT 50). After that, B cell, T cell, macrophase and NK-cell rate in the abdominal cavity were evaluated by FACS analysis.
Results: 1. Cytotoxicity of SAKT was low on WB-F344 cell lines. 2. SAKT of 200㎍/㎖ concentration was significantly effective in the restoration of GJIC(p<0.01). However, effects of SAKT both of 400㎍/㎖ and 800㎍/㎖ concentration were not significant. 3. In the SAKT 10 group, T cell(p<0.001), B cell(p<0.01) and macrophage (p<0.001) were significantly increased, but the increase of NK cell was not significant. In the SAKT 50 group, T cell, B cell, macrophase and NK cell were not significantly increased.
Conclusion: According to the above results, It is assumed that SAKT is useful for the prevention of tumorigenesis and for cancer treatment. |
