| ABSTRACT |
Objective :
Our study was carried out to identify anti-cancer effects of Banmyo-tang, a water extract on the human stomach cancer cells.
Methods :
In order to observe the anti-cancer effects and mechanism of Banmyo-tang, a water extract, ether the extract and methanol extract of mylabris phalerata on human stomach cancer cells, they were administered to human stomach cancer cells and mitochondria derived from rats s liver. MTT assay was conducted to compare and examine the change of cytostaticity in stomach cancer cells with time, and apoptosis assay was performed to examine its effects on apoptosis by using DAPI dye and counting the number of cells that developed in an apoptotic body. In addition, the quantitative RT-PCR was used to examine their effects on the revelations of bcl-2, bax and p53, genes related to apoptosis.
Results:
In MTT assay, the groups, administrated with the extract and methanol extract had no significant cytostaticity, but groups administrated with Banmyo-tang and water extract had significant cytostaticity statistically. Apoptosis assay demonstrated that groups of administrated with the ether extract and methanol extract didn t show an apoptosis increase significantly, but to groups with Banmyo-tang and water extract showed apoptosis increase in stomach cancer cells.
Conclusion:
Banmyo-tang and water extract showed an increase of cytostaticity and apoptotic inducement of human stomach cancer cells by decreasing the mitochondrial membrane potential and increasing the permeability transition. |
