ABSTRACT |
Objective : This study was carried out to investigate effect of Dodamtang(DDT) on inducible NO synthase (iNOS) level in lipopolysaccharide(LPS)-activated endothelial cells.
Methods : We observed effects of DDT on the levels of iNOS and its related inflammatory mediators. These were studied in LPS-stimulated human umbilical vein endothelial cells.
Results: DDT, at concentrations up to 5 mg/ml, did not exert any cytotoxity assessed by dye exclusion ability assay in control or LPS-activated cells. DDT significantly reduced NO production in LPS-activated endothelial cells, whereas it did not affect the basal NO level in control cells. Western blot analysis revealed that DDT suppressed the expression of iNOS in LPSactivated endothelial cells. In contrast, it did not affect the constitutive endothelial NOS (eNOS) level. DDT significantly reduced the release of tumor necrosis factor (TNF-α ) in LPS-activated endothelial cells. Electrophoretic mobility shift assay indicated that DDT down-regulated the activation of nuclear factor kappa B (NF- κB) in LPS-activated endothelial cells.
Conclusions: These results suggest that DDT could suppress the action of inflammatory mediators in endotoxin-induced vascular lesions by inhibition of NF- κB activation and iNOS expression. |
